Centrifugation
Centrifugation can lead to
• a reduced longevity of motility (Alvarez et al., 1993)
• affected DNA integrity (Zini et al., 1999; Aitken et al., 2014).
Always use g force NOT rpm!
Calibrate the centrifuge; to achieve the correct g force, use the
equation: Rpm = √[ (g/(1.118 x r)] x 103
For example; to achieve 300 x g when radius = 165 mm the centrifuge speed must be: Rpm = √[ (300/(1.118 x 165)] x 103 = 1275 http://drycake.com/calculator/gforce.php
300g – 500g is recommended

Incubation time influences sperm quality
‐  progressive molity (Anbari et al., 2016)
‐ DNA fragmentaon over me ( 1 hours) at RT and 37C (Anbari et al., 2016; Nabi et al., 2014; Zhang X et al. 2011)
‐ Samples with elevated DNA fragmentation levels are more susceptible for DNA fragmentation (Rougier et al., 2013)
