Sperm preparation in the laboratory
Discontinuous density gradient centrifugation
Pros
• High recovery rate
• Clean fraction is obtained
• Density gradient will effectively remove
Lymphocytes, epithelial cells, abnormal, senescent and immature sperm, cell debris, seminal fluid, bacteria and to some extent viruses, sources of ROS, sperm with abnormal DNA
• The amount and volume of the layers can be adapted according to the initial sperm concentration, presenece of abnormal forms and motility (Nijs & Ombelet, 2003; Nijs, 2009) – very low sperm density samples can be processed

• Time consuming • More cost
• >45 minutes
