Sperm preparation in the laboratory
Discontinuous density gradient centrifugation
• This method uses centrifugation of seminal plasma over density gradients consisting of colloidal silica coated with silane, which separates cells by their density.
• A simple two‐step discontinuous density‐gradient preparation method is prepared, typically with a 40% (v/v) layer over an 80% (v/v) density lower layer.
• Place 1 ml of semen above the density‐gradient media (More than one tube per semen sample may be used, if necessary.)
