Discontinuous density gradient centrifugation
• Centrifuge at 300–400g for 20–30 minutes.

Discontinuous density gradient centrifugation
• Remove most of the supernatant from the sperm pellet.
• Take a new pipet and bring the pellet in 5 ml of supplemented medium and resuspend by gentle pipetting (to aid removal of contaminating density‐gradient medium) and centrifuge at 300g for 5‐10 minutes.
• Repeat the washing procedure
• Resuspend the final pellet in supplemented medium by gentle pipetting
