Sperm preparation in the laboratory
Swim‐up technique
• Gently return the tube to the upright position and remove the uppermost 1 ml of medium with a pipet. This will contain highly motile sperm cells.
• Bring in tube with 1.5–2.0 ml of supplemented medium.
• Centrifuge at 300–500g for 5 minutes and discard the
supernatant.
• Resuspend the sperm pellet in 0.5 ml of supplemented medium

Pros
• Easy
• Cost effective
Swim‐up technique
• Usually recovery of a very clean fraction with highly motile spermatozoa
Cons
• Only to be used for high concentration sperm samples with motility
• Low yield
• ROS is present in the pellet oidave damage
