Swim‐up technique
• Spermatozoa may be selected by their ability to swim out of seminal plasma and into culture medium. Hence “swim‐up” technique.
• The direct swim‐up technique can be performed either by layering culture medium (1,2 ml) over 1 ml of liquefied semen or by layering liquefied semen under the culture medium
• Incline the tube at an angle of about 45°, to increase the surface area of the semen–culture medium interface, and incubate for 1 hour at 37 °C.
