Sample collection
Abstinence
‐ WHO: World Health Organization (WHO) recommends collection of semen samples after 2–7 days
‐ Time (Days) influences parameters such as
• semen volume 
• sperm concentration 
• DNA fragmentation (↑ or )
‐ A short (24‐hour) abstinence period negatively influences chromatin
quality (immature DNA packaging)(De Jonge et al., 2004)
‐ Very short time interval production (2 hours) influences (Mayorga‐Torres et al. 2016)
• semen volume ↓
• sperm concentration ↓
• DNA fragmentation ↑ or 
• Total mole count ↓
Advised: abstinence interval of ≤ 2 days

Mode
‐ On site in dedicated facility
‐ Method: masturbation or use of silastoc condom
‐ Health situation (fever, medication, drugs, ...)
‐ Sterile container, wide mouthed , cytotoxicity tested (Nijs et al., Weight defined (WHO 2010)
‐ Signature for confirmation of ID patient
‐ Transport to laboratory at body temperature
‐ Double witnessing in place
‐ Process asap after liquefaction ≥20 minutes
