TAKE HOME MESSAGE
• Cell therapy using autologous peripheral blood CD133+ BMDSC offer a safe and efficient therapeutic approach for patients with refractory AS and/or (EA) wishing to conceive.
• Our primary objective was the reconstruction of the endometrium which occurred in 15 out of 16 patients assessed by histeroscopy, histology and US. Mature vessel density increased as well as duration and intensity of menses returning to the initial levels 6 months after the treatment.
• The secondary objective was the functionality of the newly formed endometrium by attempting to conceive. Several pregnancies were achieved spontaneously and through ART after cell therapy. We have 5 babies born. Nevertheless, conception success remains to be improved.
• The injection of human SPIOs labeled CD133+ BMDSCs in a murine model of AS confirms that these cells engraft around endometrial vessels, at low rates (0,60% of injected cells) inducing proliferation of surrounding cells through paracrine molecules such as thrombospondin 1 and insulin-like growth factor 1.
