Next Generation Sequencing (NGS)
Concept
- Next-generation sequencing (NGS) or high-throughput sequencing, is the catch-all term used to describe a number of different modern sequencing technologies
- Allow to sequence DNA and RNA much more quickly and cheaply than the previously used Sanger sequencing (revolutionized the study of genomics and molecular biology)
Technologies
- 2005: Roche Life Sciences first company commercializing a NGS platform
- 2006: Illumina (Solexa) sequencing-by-synthesis technology → Miseq
- Others: Ion Torrent (PGM, Life Technologies) and PacBio (Pacific Bioscience)
Main advantages
- Increased number of sequenced bases per run
- Parallel sequencing of different target regions
- Clonal sequencing of single DNA molecule
