Prospective study-72 patients. Individual culture till PN (30μl), groups; 30 μl/emb., or for 3-4 embryos Group culture superior for compaction, blastulation, blastocyst quality, and more life births
Prospective-78 patientsIndividual culture (20 μl) from day 1-3, A. Till day 5 randomly grouped (2-5 embs./50 μl) regardless of embryo quality,B. Good/poor quality grouped, group B type sig.promoted blastocyst development, utilization rate.
No diff. in IR&PR
Randomized comparitive study , individual culture1st. 88 patients; Large culture volume( 35μl)vs. small(15μl), 2nd.
77 patients;large vs. mini-drop (7 μl) . 1st. No sig. diff. for blastocyst formation and embryo quality till blastocyst, 2nd. No diff.till day 3 but sig. higher blastocyst formation for 7 μl
Prospective sibling oocyte study- 23 patients, individual culture either 25 or 7μl. No sig.for fertilization and day 3 embryo Quality, but sig. less cell no. for 7μl., sig. higher blastocyst formation and good quality, utilization rate/oocyte for day 3 & blastocysts for 25μl.
Incubation Volume/Embryo Grouping
• Still no consensus on the incubation volume, or the ideal culture way; individual or in groups?
• With the introduction of novel technologies like PGT, or time-lapse incubation more and more clinics move to individual culture
• In order to culture in reduced volumes( 20-50μl.)an oil overlay is required to prevent the evaporation of media, thereby reducing harmful effects of increases in osmolarity, and reduces changes in pH caused by a loss of CO2 from the medium when culture dishes are taken out of the incubator for embryo examination
